The half-life of tadalafil, when taken orally, is approximately 17 hours. That results in increased blood flow into the penis and smooth muscle relaxation, leading to an erection.

Avanafil

The mechanism of action begins with a drug-drug interaction that leads to inhibition of phosphodiesterase type 5  (PDE5), which is responsible for cGMP degradation in the corpus cavernosum. Avanafil inhibits PDE5, resulting in increased levels of cGMP and greater smooth muscle relaxation. This effect increases blood flow into the penis during sexual stimulation, thus increasing erection size and duration. In addition, avanafil also relaxes penile helicine arteries and trabecular smooth muscle cells. Increased inflow of blood provides more oxygen to the cells in the penis, which may increase penile size.
